    Unusual upright spinning disk confocal built for zebrafish, drosophila, and brain slice work, with both 1-photon and 2-photon holographic photostimulation via spatial light modulators. Phasor-based patterned illumination makes it well suited to optogenetics and photo-uncaging.

    A spinning disk confocal on an upright stand, well suited to zebrafish, drosophila, explants and brain slice work. It is one of the few upright spinning disks anywhere, and it carries both 1-photon and 2-photon optical stimulation devices with Phasor holography.

    Configuration. Spinning disk excitation at 488 and 561 nm, with holographic excitation at 405, 488 and 561 nm plus a two-photon path. One-photon photostimulation runs at 405, 477 and 594 nm through the SLM; two-photon photostimulation covers 780 to 920 nm through the same. Phasor patterned illumination is what makes the optogenetics and photo-uncaging work practical. Transmitted-light brightfield runs through the spinning disk. Z-stacks, time lapse and live imaging are all supported, with live work aimed at slices and small organisms such as zebrafish, drosophila and explants.

    Objectives. 5x; W Plan-Apochromat 20x/1.0 DIC VIS-IR M27 (water, WD 1.8 mm); 40x.

    Software. Slidebook 6, with Slidebook Reader available as a free viewer. Laser use authorization 4149.
