    The McGill Primary Airway Cell Biobank (PACB) offers a comprehensive collection of high-quality human primary airway cells and specimens for respiratory disease research. As the first facility of its kind in Canada, the PACB provides standardized biological materials from both normal donors and those with various airway diseases, including Cystic Fibrosis (CF), COPD, asthma, chronic bronchitis, and emphysema. These resources are crucial for advancing studies in airway physiology, pathophysiology, cell biology, inflammation, mucosal immunity, and translational research aimed at developing new therapeutics. The biobank's extensive inventory includes a variety of primary airway tissues and cells. Available tissue types encompass trachea, superior bronchus, inferior bronchus, smooth muscle, parenchyma (alveolar tissue), and endothelium. These tissues are provided as sections from fixed tissue in paraffin blocks or OCT, as well as snapfrosts of fresh tissues. Cellular offerings include cryovials of 1 million frozen, viable primary Human Bronchial Epithelial (HBE) cells, highly-differentiated HBE cells on porous filters (transwells/snapwells), and HBE cells cultured on plastic flasks (T25). Other available specimens include Human Tracheal Gland (HTG) cells, airway fibroblasts, airway smooth muscle cells, sputum, nasal brushes, nasal polyps, and nasal internal turbinate. Specialized cell culture media, such as BEGM and ALI media, are also available. PACB-distributed cells are considered the "gold standard" for in vitro studies, closely mimicking the in vivo airway epithelium, making them superior to immortalized cell lines for testing hypotheses related to disease pathogenesis and preclinical drug development. The biobank ensures rigorous quality control, with all preparations safety tested by an independent diagnostic lab for mycoplasma, bacteria, yeast, fungi, HIV-1, HIV-2, Hepatitis A, B, C, and Hantavirus. Verification processes include assessment of cell purity, viability, capacity for differentiation into pseudostratified mucociliary epithelium, and transport function via Ussing chamber assays. The PACB is a non-profit facility certified by the UBC Office of Biobank Education and Research Biobank Certification Program, affiliated with the International Society for Biological and Environmental Repositories (ISBER). Detailed donor information is provided for each lot of primary HBE cells, including sex, age, BMI, diagnosis, cause of death, medication, antibiogram, serology, and FEV1. For CF donors, specific mutation and CFTR genotype validation are also performed. This comprehensive data supports robust and reproducible research outcomes.
