    Paraffin Tissue Processing and Embedding

    Lady Davis Institute
    Core facility
    Jewish General Hospital

This service involves the meticulous preparation of tissue samples for microscopic examination by embedding them in paraffin wax. The process typically includes several steps: fixation (often with formalin) to preserve cellular structure, dehydration through a series of increasing alcohol concentrations to remove water, clearing with a solvent like xylene to remove alcohol, infiltration with molten paraffin wax, and finally, embedding the tissue in a solid paraffin block. This technique is fundamental in histopathology and research, providing structural support for thin sectioning, enabling long-term storage of tissue blocks at room temperature, and allowing for various staining procedures. Formalin-fixed paraffin-embedded (FFPE) tissues are widely used for disease diagnosis, studying tissue morphology, and research applications.
