    TIRF/Spinning Disk – Total Internal Reflection Fluorescence Microscope

    McGill University

    Total Internal Reflection Fluorescence (TIRF) microscope for looking within ~100 nm of the microscope slide. Specifically well suited for endocytosis, exocytosis, adhesion and membrane protein/lipid studies.

    Features:

    • Total Internal Reflection Fluorescence (TIRF)
    • Variable pinhole Spinning Disk Confocal
    • Rapid simultaneous two colour imaging on EMCCD
    • Environmental Control for live cell imaging


    Light Sources:

    • Halogen Lamp for Transmitted Light
    • Wide Field Microscopy - X-Cite 120 LED (370-700nm), 100W
    • Diode Laser - 488nm - 100 mW - with FLCRtm control system
    • Diode Laser - 561nm - 100 mW
    • Diode Laser - 642 nm - 110 mW


    Objectives:

    • 10X N-PLAN, NA=0.25
    • 20X HC PLAN APOCHROMAT, NA=0.70,
    • 63X HCX PLAN APOCHROMAT, NA=1.47, OIL, Corr, TIRF


    Fluorescence Emission Filters (TIRF) - separate widefield cubes are also avaliable.

    • EGFP (ET 525/50nm)
    • mCherry (ET 620/60nm)
    • EGFP/mCherry Dual Cube

    Fluorescence Probes:

    • EGFP
    • mCherry
    • Cy5/IR-FP

    Detectors: (sCMOS are in place but can be switched for EM-CCD cameras depending on the application)

    • Hamamatsu ImageEMX2 EM-CCD - 2 Cameras
    • Hamamatsu C13440-20CI sCMOS - 2 Cameras

    Additional Features:

    • DIC Optics
    • High resolution sequential multi-colour imaging


    Location:

    • Located in Bellini Building, Room 147.

    Initial training sessions usually last about 3 hours and include the hourly microscope fee plus the technical support fee.

    IMPORTANT: ALL training and technical support is by appointment ONLY and will depend on the availability of the ABIF staff.
